Das Blut von Dawnwalker spielt Mitte des 14. Jahrhunderts in einer alternativen Version Europas und folgt der Reise eines jungen Mannes namens Coen. Nach den dramatischen Ereignissen, die im Intro dargestellt wurden, erhält Coen übernatürliche Kräfte und beginnt eine Mission, um seine Lieben vor den Klauen der Vampire zu retten. Die Dringlichkeit seiner Suche wird durch eine enge Frist unterstrichen - Coen hat nur 30 Tage und Nächte, um sein Ziel zu erreichen. Die Zeit im Spiel verläuft in bestimmten Momenten der Spiele, in der die Spieler es akribisch verwalten müssen.

Während seiner Reise sieht sich Coen erhebliche moralische Dilemmata gegenüber und entscheidet, ob er sich an seiner Menschlichkeit festhalten oder seine aufstrebende vampirische Natur annehmen soll.
